Abstract (EN)

Goal: Juvenile idiopathic arthritis (JIA) is a chronic, inflammation-induced textural erosion that is not well understood by the pathogenesis stages. There are many clinical and laboratory criteria in the diagnosis of disease. Among the changes that occur in the pathogenesis of JIA are migration of synovial fluid in groups of immunocytes. This migration leads to dislocations in mechanisms that play a role in self-regenerating touch. Molecular changes in polarity of immunocytes that migrate to both synovial and synovial fluid are discussed as important for such immunological damage. Intracellular polarity is controlled by WNT signaling pathways. Proteins involved in non-canonical signaling pathways DMP1 proteins involved in the pathways PARD3, DVL1, SCRIB and Ca+2 control the shape of cells, their connections to each other, their formation in a stable state, and the mechanism of migration due to signal from the extracellular domain. In this study, we aimed to investigate the localization of cell polarity in JIA pathogenesis and to investigate the pharmacological modulation of this change, by determining gene expression levels to evaluate the functional changes of the proteins involved in cell polarity in the synovial stem cell damage mimetic model. Materials and Methods: Implications of JIA damage for this purpose In order to demonstrate the polarity change and pharmacological modulation by immunostimulants in the synovial root cell culture model, expression levels and in-vitro drug effects of important proteins of the wild-type WNT pathway were evaluated. In experiment, 3 cells of 6 wells were cultured with Sinovial stem cells. A total of 18 well, 3 wells were manipulated in the form of 2 passages with a 48-hour passage through the normal culture as 48 wells, respectively, as well as IL-1, TNF-a, IL-1 + TNF-a, IL-1 + Canakinumab, TNF-a + Adalimumab It was. Then, mRNA isolation was performed from the samples taken from the culture medium. After cDNA was obtained by reverse transcriptase PCR from isolated mRNAs, cDNAs were amplified using ABI7000 RT-PCR as expression levels Ct value using PARD3, DVL1, SCRIB, DMP1 Taqman prepared probes previously ordered with RealTime PCR Was measured in the device. The results were evaluated by Ct. V Results: According to the evaluations, it was observed that the intracellular polarity pathways were changed in JIA damage imitations and pharmacological modulations. A decrease in gene expression was observed in IL-1 and TNF-a impairments that would result in reduced polarity proteins in the environment. In addition, the level of expression was enhanced by mimicking DMP1 protein damage, which is the initial mineralization at the time of damage, which protects the formation in a stable state. The changes in expression levels in the pharmacological modulations were narrowed but observed only in the Dvl1 gene encoding the DVL1 protein, which is also completely stopped in the cell, which also helps other life pathways. Discussion and Method: According to the results obtained in our findings, the JIA frame damage is lost due to internal damage to stem cells, due to internal damage to the cell polarity and indirectly to the cell-cell junctions and consequently, the regeneration ability of the stem cells is decreased due to binding / polarity loss, It was concluded that the measures worked better.
