ABSTRACT dummy The Effects of Circadian Clock Genes On The Formation of Benign and Malignant Brain Tumors Bilge Onur ŞİŞ Master's Thesis FIRAT UNIVERSITY Graduate School of Natural and Applied Sciences Department of Biotechnology July 2023, Pages: x + 32 Circadian clock genes are genes that play an important role in regulating the biological clock of the organism. Circadian clock genes ensure the harmonious functioning of biological processes in the body and influence many processes, including sleep-regulation, metabolism, and behavior. The circadian clock is a feedback loop consisting of four genes/proteins called BMAL1, CLOCK, Cyrptochrome and Period. Studies show that disruption of the circadian clock plays an important role in the development of malignant tumors, including gliomas. However, the molecular mechanisms of the genes controlling the circadian rhythm in the formation of benign and malignant brain tumors have not been investigated. Therefore, it is very important to investigate the molecular mechanisms of BMAL1, CLOCK, Rev-Erbα and Per2 biological clock genes, which are thought to be effective on brain tumor formation. When the results were analyzed, a strong negative correlation was found between ACTB and Clock clock gene levels, and a strong positive relationship between ACTB and Bmal1 and Rev-Erbα gene levels (p<0.01). In addition, it was determined that there was a significant positive correlation at p<0.01 level between Bmal1 and Clock gene and Per2 gene, and a significant positive correlation at p<0.001 level with Rev-Erbα. Again, a strong positive correlation was found between the Clock gene and the Per2 gene at the p<0.001 level. When the correlation table between blood biochemical findings and gene levels was examined, it was determined that WBC values showed a significant positive correlation at the p<0.05 level with Bmal1 and Rev-Erbα genes, and at the p<0.01 level with the Clock gene. It was determined that CRP values showed a significant positive correlation with the Clock and Per2 genes at the p<0.05 level, and with the Bmal1 gene at the p<0.01 level. In conclusion, it is to contribute to the understanding of the role of these genes in brain tumorigenesis and to gain more insight into the influence of circadian rhythm on tumorigenesis.
