Abstract (EN)

It is one of the most important complications that cause serious morbidity and mortality due to infections that occur in cancer patients, the tumor itself, neutropenia or anti-neoplastic treatment (cytotoxic, immunosuppressive, etc.). Staphylococcal infections are the most common bacterial infections in acute myeloid leukemia (AML), which constitutes 15% of childhood acute leukemia and 50% of deaths from leukemia. Enterotoxin type A (SEA), one of the most effective toxins of Staphylococcus aureus (S. Aureus), is a virulence factor that causes food poisoning, toxic shock syndrome, pneumonia and infections like septic shock. Although there are some studies on the effects of SEA on cancer cells, these studies have mostly been done on solid tumors. There are very few studies examining the effect and mechanism of action of SEA on blood cancers, especially acute leukemias. This study aims to investigate the effects (inhibiting / activating effect) of SEA on 6 different AML and a control cell line and the molecular mechanisms of these effects; It also aims to examine the effects of SEA on resistance to some chemotherapeutics frequently used in AML and its mechanism of action responsible for it. The effect of SEA on AML cell lines will be investigated by applying proliferation, proliferation, transwell migration and invasion tests in cell lines after treatment with SEA. In addition, in silico analyzes will be made to determine the genes and pathways that may be responsible for the effect of SEA. FDA approved idarubicin and cytarabine drugs, which are frequently used in AML treatment, will be used to determine the resistance profiles of SEA's cell lines against chemotherapeutic drugs; The effects of SEA on the sensitivity or resistance of AML cell lines to these drugs will be determined by cell viability tests. It is believed that understanding the molecular basis of the relationship between cancer-associated infections and cancer processes will bring new approaches to the treatment of cancers, especially leukemia. It is also important to enlighten the effects of the aforementioned infections on drug resistance developing in AML and the molecular mechanism of these effects. The data of this study will lead to future in vitro and in vivo drug development / targeting studies.
