Abstract (EN)

In this thesis, it is aimed to realize the domestic production of biological indicators, which are sterilization process control kit which is not obliged to be imported in Turkey. Preparing stocks of selected microorganism spores which are used as biological indicators consists of the steps of developing with appropriate procedures, culturing, spore production, purification and maintenance of spore life. Stock spore suspensions consist mostly of passive and non-proliferating spore forms which are maintained in a nutritious environment. The variables optimized for determination of optimum growth and sporulation medium in liquid culture of Bacillus atrophaeus ATCC 9372 are soybean flour, corn steep liqour, yeast extract, stock mineral solution, temperature, pH, ZnSO4, K2HPO4, KH2PO4 and glucose. In the screening study conducted with the Design Expert 7 program by Placket Burman method, 11 variables were evaluated. It was decided to optimize the variables of soybean flour, corn steep liqour, yeast extract, and stock mineral solution. For the response surface method (RSM), the factorial central composite design (CCD) with 6 repetitions at the center was considered appropriate. According to RSM results, the program yielded 39.8 g / L soybean flour, 30 g / L corn steep liqour, 1 g/ L yeast extract and 1mL / L stock solution formulation optimally. The number of spores obtained was 4.1x108 spores / mL and was sufficient for the production of biological indicators.
