Abstract (EN)
In this study, the phytase enzyme from Ochrobactrum anthropi isolated from wastewater treatment plant was produced and characterized. The culture conditions were optimized for maximum enzyme production. The best carbon and nitrogen sources for maximum phytase production were 1% maltose and 1% yeast extract, respectively. Moreover optimal pH 8.5, temperature 37oC, agitation rate 300 rpm, inoculum size OD600: 0,06, inoculum age 9 hours were found. Molecular weight of intracellular phytase enzyme which produced under the optimal conditions from Ochrobactrum anthropi, was estimated approximately 22 kDa and 13 kDa on SDS-PAGE after the purification with gel-filtration chromatogphy. The optimum activity of intracellular enzyme was emerged 40oC and pH 5.4. Enzyme activity was increased to 117 % after the pre-incubation at 90oC 30 minutes. Enzyme activity significantly inhibited with 1 mM Triton X-100 (91%), Tween-20 (56%) ve Tween-80 (55%) and stimulated with 1mM NaCl (104%), MnSO4 (105%), MgSO4 (102%), gliserol (106%), EDTA (103%), Asetik asit (117%), 2-merkaptoetanol (109%). As a result, it is seen that due to the characteristic features of the Ochrobactrum anthropi enzyme that can be predicted that result of feed addition at monogastrics becomes prevention of environmental pollution.
