Investigation of antioxidant parameters of sulforaphane on the Caco2 cell line

Abstract (EN)
Cancer is a disease in which cells grow uncontrollably and spread to other parts of the body. Various chemotherapeutic drugs have been developed for the treatment of cancer and used for the treatment of tumours. Drugs such as chemotherapy and radiotherapy used for therapeutic purposes cause negative side effects such as weakening of the immune system and hair loss while fighting cancer cells. Therefore, the tendency towards medicinal plants with anticarcinogenic effect has increased. The effects of sulforaphane, which is found in cruciferous family such as broccoli and cabbage among medicinal plants, on many diseases such as cancer are being examined. The aim of this thesis is to investigate the cytotoxic and antioxidant effects of sulforaphane on human colon cancer cell line Caco2 cell line. The 24 h inhibition value (IC50) of sulforaphane (SFN) on Caco2 cell line was determined as 181.795 µM by MTT assay method. Cells were exposed to SFN concentrations of 1/10 and 1/100 of this value (18.2 µM and 1.82 µM) for 24 hours. After exposure, glutathione, advanced oxidation protein products and malondialdehyde tests were performed to determine the oxidative stress and antioxidant effects of sulforaphane on cells. A significant increase was observed in glutathione values of control groups compared to 1.82 µM SFN groups and 18.2 µM SFN groups showed a statistically significant increase compared to 1.82 µM SFN groups (P<0.05). It was observed that sulforaphane decreased advanced oxidation protein products and malondialdehyde levels in Caco2 colon cancer cells. As a result of this thesis study, it is seen that SFN exposure of Caco2 cells has a cytotoxic effect and changes in glutathione values
