Abstract (EN)

In this study, the profile of the microbial community inhabiting sediment layer of Tuz Lake was revealed by the 16S rDNA gene-targeted Illumina MiSeq high-throughput sequencing, gene cloning and culture studies. Metabolic potentials and environmental roles of microorganisms in this environment have been demonstrated by metagenomics, single cell genomics analysis and culture dependent EcoPlate studies. In the data obtained by cloning and MiSeq sequencing, it was detected that the phylotypes related to Halobacteriales, Thermoplasmatales, "Methanonatronarchaeia", Methanobacteriales, "Rhodothermales", Bacteroidales, Halanaerobiales, Gemm-4 (Gemmatimonadetes), KB1 / OP1 (Acetothermia), Desulfobacterales, Desulfovibrionales and Oceanospirillales groups were highly represented. General characteristics and metabolic potentials of 25 draft genomes obtained by metagenomic analysis has been revealed. These genomes are phylogenetically related with Halomonas (2), Halodesulfurarchaeum (1), Thiohalorhabdus (1), Rhodobacteraceae (1), Halanaerobiaceae (2), Lentimicrobiaceae (2), Chloroflexi (1), Candidatus Izimaplasma (1), Bacteria domain (3), Ca. Marinimicrobia (2), Ca. Omnitrophica (1), Ca. Microgenomates (1), Candidate divsion MSBL-1 (3), Candidate division SA1 (1), Ca. Bathyarchaeota (1) and Ca. Nanohaloarchaea (1). Approximately half of these draft genomes appear to belong to groups without any cultured representatives. In some sediment samples, Ca. Acetothermia phylotypes were represented at a high rate (~ 30%). In the single cell genomics study, 6 single cell genomes of Ca. Acetothermia, which were determined as the target group, were analyzed and genome characteristics and possible metabolic profile of these representatives were revealed.
