Abstract (EN)
In this study, production conditions for xylanase enzyme production in solid culture fermentation were optimized. Various agroindustrial wastes (wheat bran, oat bran, rice bran, corn kob, sawdust, malt sprout) were used as substrate for production of xylanase from Aspergillus niger. Effect of substrate, inorganic nitrogen source, amount of inorganic nitrogen source and initial moisture content (%) to xylanase production were determined. Maximum enzyme activity (759.15 U/g dry substrate) was obtained at optimum conditions (malt sprout, diammonium hydrogen phospate added mineral salt solution, initial moisture ratio 70%). Inoculation ratio and aeration flow rate were studied for xylanase production in tray bioreactor. Under optimum conditions (0.1 ml spore suspension/g substrate inoculation, 2.5 L/min aeration flow rate) xylanase activity was determined as 1312.04 U/g dry substrate. Keywords: xylanase, malt sprout, Aspergillus niger, solid state fermentation
