Abstract (EN)
Obesity, It is a health problem that results in excessive fat storage in the body if the energy taken is more than the energy given, and accordingly, it is a health problem that progresses with the risk of many chronic diseases, including type 2 diabetes, insulin resistance, cardiovascular diseases and cancer. Changes in the microenvironment in obesity increase the risk of breast cancer, and the increase in obesity is in a positive correlation with breast cancer. The prevalence of obesity is increasing in our country and in the world, and in this case, it leads researchers to find alternative treatment methods. These alternative treatment methods are largely composed of medicinal plants. Urtica dioica L. is a plant that has been considered medicinally valuable since ancient times. Thanks to the active components in the plant, it is included in many studies in the literature due to its anti-inflammatory and anti-cancer properties. In this study, 3T3-L1 adipocytes and LPS-induced control groups were used to observe the effect of Urtica dioica on 3T3-L1 adipocyte cells. LPS has been used as a factor that triggers increased inflammation. In order to examine the regulation at the transcriptional level, RNA was obtained from all groups, and then the RNAs were converted to cDNA and BRCA-1, BRCA-2, Her-2, TNF-α, IL- 6, Transcriptional expression of IL-2, MCP-1, DGAT-1, Fasn, LPL primers was examined. qPCR was used for analysis and normalization of the expression of the genes provided 18S, the housekeeping gene. As a result of the study, a decrease in the expressions of BRCA-1 BRCA-2, LPL and Fasn, MCP-1, DGAT-1 genes was observed and these changes were found to be statistically significant. Then, the active components of the plant dissolved in ethanol were found from the literature. Ligands were prepared using these active ingredients from Pubchem. The proteins of the genes we found significant were found in PDBQT format. Molecular docking of ligands and proteins was performed.
