Effect of exopolysaccharide (EPS) production by Lactobacillus spp. originated from vagina on antibiotic susceptibility and adhesion on cell surface

Abstract (EN)
In this study, exopolysaccharide (EPS) production and hemagglutination (HA) capability of 57 Lactobacillus spp. strains originated from human vagina were investigated. The adhesion capabilities of these strains were measured by using gel HA technology test in vitro. EPS productions of strains were determined as a range of 20?368 mg/L. In addition, 77.2% of the strains showed high HA capability (+4, +3), %12 of the strains showed no hemagglutination. Lactobacillus crispatus G10 showed maximum EPS (368 mg/L) production and also possessed high HA (+4) ability. Lactobacillus acidophilus I4 strain which produced lower EPS (30mg/L) did not show hemagglutination. Antibiotic resistant Lactobacillus spp. against 12 antibiotics were determined by disc diffusion method and results were evaluated according to the NCCLS criteria. Lactobacillus strains were resistant to gentamicin (100%), ofloxacin (98%), ciproflaxacin (96%) and streptomycin (88%) and sensitive to doxycicline (100%), chloramphenicol (98%), tetracycline (91%) and eritromycin (89%). Minimum inhibition concentrations of Lactobacillus spp. were determined as a range of 1000-8000 ?g/ml concentrations of metronidazole and 64?1750 ?g/ml concentrations of flucanazol. L. crispatus G10, L. gasseri H14, L. oris A2 were showed highest resistance to both metronidazole and flucanazol. Shelf life of vii these strains (L. crispatus G10, L. gasseri H14 L. oris A2) was determined at glycerinated gelatin ovules containing metronidazole and flucanazol. Also, ascorbic acid was used as excipient at ovule preparates and it was determined that ascorbic acid significantly increased the survival of all these strains. The highest survival was observed for Lactobacillus gasseri H14 in all the formulations.
