Abstract (EN)

In this study, it was aimed to determine the resistance of 22 melon genotypes collected from Van Lake Basin, 3 resistant genotypes and 2 commercial varieties to ZYMV (Zucchini Yellow Mosaic Virus) by using mechanical inoculation method, also it was aimed to determinate the presence of resistance markers ZYMV (Zucchini Yellow Mosaic Virus), CMV (Cucumber Mosaic Virus) and WMV (Watermelon Mosaic Virus) genotypes in melon genotypes and varieties through SCAR and CAPS marker methods. After mechanic inoculation disease severity was determined according to 0-5 scale by performing morphological observations in genotypes. In second stage of the study the presence of markers related to resistance genes against to ZYMV, CMV and WMV were defined in these genotypes by screening with 7 primers including 3 SCAR primers and 4 CAPS primers. While bands were obtained from SCOPE14541 primer in 21 genotypes SCAPB051046 primer that was developed based on the Creb-2 resistance gene gave bands only in the U6, U13 and YYU6 genotypes from 25 genotypes. VirSq-F19 primer that was developed based on the resistance genes to ZYMV and WMV gave band in all genotypes exluding YYU47. Only two of the 4 CAPS primers associated with the Zym resistance gene produced band. The desired gene region was amplified with the primers dCAPS-G99A and CAPS-T86C, but no restriction was performed at the desired band intervals for the restriction enzymes (Dra I / Mn II).
