Abstract (EN)

Staphylococci with microbiota elements in humans are the most important factors of community and hospital central infections. Glycopeptide antibiotics are used in methicillin-resistant Staphylococci strains. Increased use of automated systems to detect methicillin resistance increased the use of glycopeptide antibiotics even in low and false MIK results. This result accelerated the development of glycopeptide resistance in cases. Increased resistance caused difficulties in the treatment of infections, which led to the investigation of the genetic basis in order to find different treatment methods. In this study, Staphylococcus strains isolated from laboratory samples were examined. Vitek 2 automated system and conventional methods, 3 of 30 strains were identified as S.aureus, 27 was identified as KNS. These strains were found to be resistant to methicillin, fusidic acid, penicillin, tetracycline, trimethoprim-sulfamethoxazole, ciprofloxacin, clindamycin, erythromycin and gentamicin by Kirby Bauer disc diffusion method. Sensitivity conditions were determined by vancomycin and teicoplanin microdilution method. In the results, the presence of mutations in the tcaRAB gene region were investigated by PCR method in strains resistant to teicoplanin. The presence of mutations by PCR was determined as 15 resistant staphylococci strains, 10 of them were methicillin resistant, vancomycin sensitive and teicoplanin resistant, 5 of them were methicillin, vancomycin susceptible teicoplanin resistant. According to PCR results, gene region in 15 strains resistant to teicoplanin were amplified. In conclusion, the effective treatment of hospital infections and the genetic basis of teicoplanin resistance in minimizing the resistance generated by mutations are important for determining the measures to be taken.
