Abstract (EN)
ABSTRACT In Vitro Comparison of Pharmacological Properties of Newly Synthesized Platinum-Based N Heterocyclic Carbene Complexes with Cisplatin Aim: To evaluate the potential of the original eight synthesized platinum N-heterocyclic carbene (Pt-NHC) complexes as leading molecules in pharmacological research and drug development studies. Material and method: The pharmacological properties of Pt-NHC complexes were evaluated on SH-SY5Y (human neuroblastoma cell line), A549 (human lung cancer cell line), MDA-MB-231 (human breast cancer cell line), and HeLa (human cervical cancer cell line) cells. Cisplatin and Pt-NHC compounds were applied to the cells at doses of 1 μM, 5 μM, 10 μM, 25 μM, 50 μM, 100 μM. The cytotoxic effects of the compounds, their effects on cell migration in cancer cells, their antimicrobial and antifungal activities, and their relationship with apoptosis were evaluated. Cytotoxic effect by MTS method, cell migration by wound healing test, antimicrobial and antifungal activities by routine activity tests, expression levels of Bax and Bcl-2 genes related to apoptosis. It was evaluated by RT-PCR method. Results: Pt-NHC compounds with different structures were found to show various degrees of antiproliferative activity on the tested cell lines. It was determined that the compounds showed antimigration effects by preventing cell migration. They were found to be antimicrobial effective against S.aureus, C.albicans, and MRSA. PCR. Conclusion: When the data obtained from the compounds were compared with cisplatin, Pt-NHC complexes significantly more effective than cisplatin were identified. We concluded that these compounds may be new candidate molecules for drug development studies. Keywords: Pt-NHC, cisplatin, apoptosis, anticancer activity.
