Multiplex analysis of 10 STR loci and detection of allele frequencies in Çukurova population

Abstract (EN)
Short Tandem Repeats (STRs) are DNA sequences consist of 2-6 base pair length blocks that repeat like head and tail in human genome. Showing difference in tandem repeat number among individuals, being amplified by PCR and being analyzed in degraded DNA samples make them ideal markers in forensic individualization and kinship analysis. In order to use STR loci in individualization and kinship analysis, first a population study must be done, and allele frequency distribution related to the population must be determined. Purpose of this study is to carry out the multiplex analysis of 10 STR loci (D10S1248, D1S1656, D2S1338, D22S1045, D19S433, TH01, D2S441, D6S1043, D12S391 and AMEL) and to study allele frequencies of these loci in Çukurova population and detection of population data. In this study, genomic DNAs prepared from samples which had been collected within the scope of another population study in Çukurova University Faculty of Medicine Forensic Medicine Department Forensic Genetics Laboratory were used. These people consisted of randomly chosen 100 volunteer individuals whom don't have a kinship. In analysis of isolated samples, Verifiler Kit which enables multiplex amplification of 10 STR loci was used. PCR products obtained after PCR reaction were conducted on capillary electrophoresis. In the study, 8 D10S1248 allele (11-18), 16 D1S1656 allele (8-21), 11 D2S1338 allele (14-25), 9 D22S1045 allele (10-18), 11 D19S433 allele (12-17.2), 6 THO1 6 allele (6-10), 8 D2S441 allele (10-16), 12 D6S1043 allele (9-21) and 14 D12S391 allele (15-25) were determined. Allele frequency distribution of 9 studied systems was found consonant with Hardy-Weinberg equilibrium. Heterozygosity ratio of loci was calculated as between 0.50 and 0.90, combined matching probability as 1.7x10-11, combined discrimination power as 0.999999 and combined power of exclusion as 0.9998. Due to high discrimination power that has been calculated for Çukurova region, it is believed that 9 STR systems together would be the valid genetic markers used in forensic analysis of this region.
