The effect of plant growth regulators applied to Achillea millefolium L. explants in vitro.

Abstract (EN)
In this thesis study, it was aimed to determine the effects of plant growth regulators applied to node, leaf and root explants of Achillea millefolium L., a medically and economically important plant, under in vitro conditions. Plants obtained from A. millefolium seeds germinated under in vitro conditions were used as explant source. Naphthalene Acetic Acid (NAA) from the auxin group and 6-Benzyl Amino Purine (BAP) from the cytokinin group, plant growth regulators, were applied to A. millefollium node, leaf and root explants, separately and in combination, at different concentrations. As a result of the applications applied to A. millefolium node explants, the longest shoot (3.7±0,3 cm) and longest root formation (7.0±0,68 cm) were detected in MS medium containing 0,1 mg/L NAA. The highest number of shoots (1.63±0.3 pieces) and the highest number of leaves (8.9±0.5 pieces) were determined in MS nutrient medium containing 0,1 mg/L NAA+2 mg/L BAP and 0,1 mg/L, respectively. As a result of the applications made to leaf explants, it was determined that the best root formation (%90,0 ±5,58) was in MS media containing 1 mg/L NAA + 0 mg/L BAP and the best callus formation (%70,0±8,5) was in MS media containing 1 mg/L NAA + 1 mg/L BAP. In applications to root explants, the highest (80.0±7.4%) root formation was obtained in media with 1 mg/L NAA and the highest (100.0±0.00%) callus formation was obtained in media with 1 mg/L NAA+1 mg/L BAP. As a result of the applications made to the node explants, the plants obtained from the MS nutrient medium containing the best 0,1 mg/L NAA were successfully transferred to the soil and acclimatized.
