Production of skin test material from Adana atmospheric isolate molds

In this study, test material was produced of regional molds for use in skin prick test and compared with standard allergens. Samples identified as Cladosporium cladosporioides, Cladosporium herbarum and Alternaria alternata were cultured on PDA solid medium for the production of single colonies and the pure colonies were produced on Czapek Dox liquid medium at 26°C for 14 days. The obtained mycelium was collected with filter paper and dried for 24 h and subjected to liquid nitrogen, ultra turrax, sonicator, freeze-thaw applications. Finally, the extracts were lyophilized for 24 h. The protein content of the extracts was determined by the Bradford Method. Extracts of each strain was fractionated by Column Chromatography. Protein bands and molecular weights of total extracts and fractions were determined by 10% SDS-PAGE analysis. The test materials produced were applied to 37 patients at the Department of Pediatric Immunology and Allergic Dieases Clinic of Çukurova University Medicine Faculty. In 5 patients for Cladosporium cladosporioides, test results were positive with both standard and produced test material, 9 patients for Cladosporium herbarum; positive with standard and test material, for Alternaria alternata in 35 patients; positive with standard and test material. It can be suggested that the test material produced according to the findings may be used clinically for diagnostic purposes.
