Abstract (EN)

Introduction and Objective: Insertion / Deletion (InDel) polymorphisms are getting more attention in forensic identification studies due to their low mutation rates and small amplicon lengths. InDels provide a new direction to forensic identification studies in complicated forensic cases and biological samples containing degraded DNA, where analysis with traditional STR loci fails. This study aims to determine allele frequencies and forensic efficacy values for the Çukurova population by genotyping the 30 autosomal InDel loci. Materials and Methods: Multiplex PCR of 30 autosomal InDel loci was performed with Investigator® DIPplex Kit following DNA isolation in blood and intra-cheek swab samples taken from 102 people. Following PCR, amplicons were carried out in automatic capillary electrophoresis and genotyped with the Genotyper v.3.2 software program. STRAF 2.0 statistical analysis program was used to calculate forensic efficacy values and POPTREE2 statistical software was used to calculate genetic distance between populations. Results: In the samples of the 102 randomly selected individuals of the cukurova population, all of the 30 indel loci studied contained 2 alleles, and the observed heterozygous value was calculated as 0.4784. None of the 30 InDel loci tested were found to be deviated from the HWE, and the observed and expected data were in balance (p>0.0016). In the linkage disequilibrium test between 30 different loci on 22 different chromosomes, no connection was found between loci (p>0.0016). Forensic efficacy value analysis showed that the combined discrimination power, and probability of combined paternity exclusion of the 30 autosomal InDel loci are 0.99999999999996 and 0.99995, respectively. Conclusion: Since the allele frequencies of 30 InDel loci are balanced and the potential of identification is significant, this data will be used as a database for statistical analysis in which the identity and kinship relations are investigated in the Çukurova population, as well as contributing to population genetic studies.
