Investigation of the relationship between scavenger receptorsand apoptotic proteins in patients with acute myocardialinfarction

Abstract (EN)

Cardiovascular diseases is a set of disorders in vascular and cardiac function such as cerebrovascular diseases, angina, atherosclerosis, coronary heart disease, myocardial infarction and heart failure. It is the most important cause of mortality and morbidity in both developing and developed countries. Scavenger receptors are cell surface receptors that bind to many different ligands and allow them to be taken into the cell. Oxidized LDL is an important ligand for many scavenger receptors. Activation of intracellular signaling pathways induced by modified LDL can cause uncontrolled lipid accumulation in different tissues and cells. Apoptosis is a form of programmed cell death. Especially foam cells formed after lipid accumulation in vessels, undergo apoptosis and this situation exceeds its regenerative capacity and causes damage to the vessels. In the light of this information in our study; it was aimed to examine how the stent treatment process affects scavenger receptors and apoptotic protein levels in patients with myocardial infarction clinic. For this purpose, patients without coronary occlusive lesion on coronary angiography were included in the control group, and the infarct group consisted of patients who had a clinical picture of myocardial infarction with a prominent coronary occlusive lesion on coronary angiography and who underwent percutaneous coronary stent implantation afterwards. Routine biochemistry and cardiac markers were measured from serum samples with a biochemistry autoanalyzer. Blood samples were taken at day 0, 3 rd day and 30 th day from patients who had myocardial infarction and Peripheral blood mononuclear cells (PBMC) were isolated with ficoll solution. LOX-1, CD36, CD68 and CXCL16 gene expressions in PBMCs were determined by qPCR method. In addition, BAX, BCL-2 and Caspase-3 protein levels in serum samples were measured by ELISA method. When the results were evaluated, no significant difference was found between the groups in the routine biochemistry values of glucose, triglyceride, cholesterol and LDL cholesterol. HDL cholesterol was lower in MI group. Cardiac markers troponin, CK-MB and creatine kinase values showed a significant increase in the MI group. However, no significant change was found in time-dependent PBMC LOX-1, CD36, CD68 and CXCL16 gene expressions between two groups and in MI group. Similarly, no significant changes were observed in BAX, BCL-2 and Caspase-3 protein expressions. The absence of a significant increase in scavenger receptor expression is probably due to the similar lipid profile observed between groups.
