Investigation of HİF1A gene polymorphisms in patients with allergic rhinitis

Abstract (EN)
Allergic Rhinitis (AR) is a disease characterized by symptoms such as nasal congestion, nasal discharge and itching, which are mediated by immunoglobulin E from the moment the allergen enters the nasal mucosa. It affects daily life negatively and is seen together with asthma over time. Hypoxia-inducible factor 1α (HIF1A) acts as an important regulatory protein in the transcription of genes involved in cell survival, proliferation and angiogenesis in hypoxic conditions in the cell. It also affects the production of many cytokines such as HIF1A, IL-8, IL-10, IL-22, TNF-α and the formation of inflammatory response. Based on these data, we aimed to investigate the relationship between C1772T and C111A polymorphisms in the HIF1A gene and AR disease. 100 adult patients with AR and 100 healthy individuals were included in our study. Genomic DNA was obtained from peripheral blood samples of individuals included in the study, and gene regions with relevant polymorphisms were amplified by PCR method. Restriction fragment length polymorphism analysis (RFLP) was performed for genotyping of relevant single nucleotide polymorphisms (SNPs) in the HIF1A gene. For the HIF1A C1772T polymorphism, the distributions of CC, CT, and TT genotypes in the patient group were 68%, 27%, and 5%, respectively, and 68%, 29%, and 3% in the control group. As a result of the analysis of HIF1A C111A polymorphism, CC genotype was determined in all of the patient and control groups. In conclusion, no statistically significant relationship was found between HIF1A C1772T and C111A polymorphisms and AR. Further studies investigating other SNPs in larger patient and control series are needed to elucidate the role of the HIF1A gene in the development of AR.
