Drug adsorption and release studies by using alginate hybrid structures

Abstract (EN)

Within the scope of the thesis, alginat microsphere and bead structures;and also hybrids of these structures with starch, polyvinyl alcohol, carboxymethyl cellulose and montmorillonite were synthesized. Characterization of the synthesized structures was carried out by using SEM and FTIR spectroscopy. The metformin hydrochloride (MF) and ciprofloxacin hydrochloride (SF) adsorption and controlled release performance of these structures were compared with each other. Aljinat microspheres showed the best adsorption performance for both drug active substances. At the optimum condition determined, aljinat microspheres were able to remove 53.79% of MF and 76.00% of SF from the aqueous medium. The adsorption data obtained for MF and SF were well described by the pseudo-second-order kinetic and the Freundlich isotherm models. In order to carry out controlled release studies, SF and MF were loaded onto the structures at the synthesis stage. The loading capacity and entrapment efficiency of MF and SF to the structures were determined. Based on these data, the release rates of the structures in the stomach and intestinal environment were calculated, and their compatibility with the release kinetic models was examined. The best results in the release studies were obtained with MMT-ALG-B. In the intestinal environment (pH=7.4), MF release was completed after 6 hours, while the release of SF was determined to be 82% after 24 hours.
