In recent years, nanoparticles (NPs) have been attracting extraordinary medical and social interest in medicine, veterinary medicine, and industries, but the safety of these NPs exposure remains unclear. In this regard, the present study demonstrates the synthesis and characterization of several antibiotics-based gold nanoparticles (AuNPs), as well as their biological applications as nanomedicine for these breast cancer cells and drug-resistant bacteria in cow's milk samples. In the research work, AuNPs were prepared with the metal precursor and antibiotics as reducing/capping agents. The formation of the AuNPs was monitored using UV-Vis absorption spectroscopy. The crystalline (11+3 nm) and particle size (9.2+1.8 nm) obtained from TEM and XRD analysis coincided with each other. In response to these AuNPs, the levels of caspases (3, 8 and 9), TLRs and TRPs were changed in SKBR3 cancer cells as compared to CRL-4010 cells, p53 was activated and nuclear factor kappa B (NF-κB) was inhibited simultaneously. In addition, the gene and protein expressions of caspases supported the data. The inverse relationship between p53 and NF-κB was found in AuNPs treated breast cancer cells. Furthermore, these AuNPs were used against drug resistant bacteria such as Staphylococcus aureus ATCC 2985 (24±1.5 mm) and Salmonella typhi ATCC 6539 (15±2.2 mm), Escherichia coli ATCC 25922 bacteria (13±1.2 mm) and Aspergillus flavus ATCC 9643 (15±1.3 mm) in cow's milk samples. Finally, it has been concluded that the study proves these Au(0) NPs are novel and can be used as an alternative nanomedicine in near future against SKBR3 cancer cells and drug-resistant bacteria and is equally extendable for the control of other reducible contaminants.
