II-ABSTRACTTHE EFFECT OF AMRINONE AND SILDENAFIL CITRATE TO BACTERIALTRANSLOCATIONIN ACUTE MESENTERIC ISCHAEMIA-REPERFUSIONINJURY.Two mechanisms are responsible in ischaemia -reperfusion injury. One of them iscell hypoxia and the other is reactive oxygen species (ROS). İn experimentalischaemia-reperfusion injury. Increase of ROS and bacterial translocation wereestabilshed. In our study we examined the effect of amrinone and sildenafil citrate toacute mesenteric ischaemia-reperfusion injury and bactreial translocation.We used 50 wistar Albino rats, which were have average 250-300 gram weight.They were divided into 5 groups. Nothing done to group 1. İn group 2 (controlgroup) 1cc saline solution was given intraperitoneally, laparotomy was done andacute mesenteric ischemia was performed. 50 mcg/kg amrinone was givenintraperitoneally to group 3 before the ischaemia. 0,05 mg/kg sildenafil citrate wasgiven intraperitoneally to group 4 before the ischeamia. 0,05 mg/kg sildenafil citrateand 50 mcg/kg amrinone were given intraperitoneally to group 5 before theischeamia. The activities of malondialdehyde (MDA), Myeloperoxidase (MPO),Glutathione peroxide (GSH-Px) and levels of ROS and Nitric Oxide (NO) in bloodwere examined. The examples were taken from liver, splein and mesenteric lenfoidnode and examined that the reproduction on classical tissue culture has occured ornot.Bacterial translocation was observed with PCR method. Samples taken fromterminal ileum were evaluated histopathologically. We compared the groups andMDA decrease and GSH-Px increase in the amrinone group were statisticallysignificant (p<0,05). The MPO decrease and NO increase were not statisticallysignificant (p>0,05). Microbiologic parameters were statistically significant(p<0,05). But histopathologic parameters weren?t statistically significant (p>0,05).The biochemical, histopathological and microbiological results were better than thecontrol group and statistically significant in the groups which were given sildenafilcitrate (p<0,05).As a result amrinone and sildenafil citrate were effective on decreasing cellinjury,ROS and bacterial translocation in acute mesenteric ischaemia-reperfusioninjury.Key word: Acute mesenteric ischaemia-reperfusion injury, bacterial translocation,ROS, amrinone, sildenafil citrate.X
