vTHE ISOLATION OF SOME PATHOGENS ANDSALMONELLA-SHIGELLA IN THE RAW MILKSOLD IN ANKARA AREA(M.Sc. Thesis)Merve ÜZÜMGAZI UNIVERSITYINSTITUTE OF SCIENCE AND TECHNOLOGYFebruary 2006ABSTRACTIn this study 100 samples of crude milk sold out in the streets at the districts ofAnkara between 06.10.2003-10.01.2005 has been used. The presence of thebacteria Salmonella, Shigella, Yersinia, Listeria and Staphylococcus, Citrobacter,Klebsiella, Enterobacter, Providencia, Proteus, Morganella has been investigatedand the live bacteria count, total coliform bacteria count and total E. coli countfor all samples were realized. The protease activity and lipase activity for all theisolated samples were conducted as well. The total mesophyll bacteria averageof the 100 crude milk samples were 1,6x106 cfu/ml , the total coliform bacteriaaverage were 1,1x106 cfu/ml, total E. coli count average was 7x 105 cfu/ml. Thetype distribution of the 115 isolated in this study is: 1 Shigella (0,86%), 9Salmonella (7,84%), 1 Listeria (0,86%), 32 Staphylococcus (27,82%), 17Citrobacter (14,8%), 13 Klebsiella (11,30%), 17 Enterobacter (14,8%), 3Providencia (2,60%), 16 Proteus (13,91%), 6 Morganella (5,21%). 19 pathogenmicroorganisms were isolated from the 100 crude milk samples. The 19 isolatedpathogen microorganisms were determined as 5% Shigella sonnei, 26% S. typhi,21% S.paratyphi, 5% L.monocytogenes, and 43% as S. aureus. The proteolyticactivities of the 19 pathogen bacteria isolated from crude milk has been studiedat 37 ºC at Calcium Caseinat Agar. 13 (68,43%) of the 19 isolated has shownproteolytic activity while 6 (31,57%) isolated hasn?t shown any proteolyticviactivity. In the study, lipolytic activities of the 19 pathogen bacteria isolatedfrom crude milk in Glycerol Tribütirit Agar were studied. 10 (52,63%) of the 19isolated has shown lypolitic activities while 9 (47,37%) isolated hasbt shownany.Science Code : 203.1.010Key Words : Raw milk, Salmonella, ShigellaPage Number : 62Adviser : Prof. Dr. Güven URAZvii
