Özet (EN)

Objective: Ankylosing Spondylitis(AS) is an inflammatory disease that pathogenesis is not fully understood. Inflammatory cytokine interleukin-17 (IL-17) is responsible for the creation of cellular response. IL-17 has six family memmbers(A,B,C,D,E and F). In this study we aimed that there can be a relationship between Ankylosing spondylitis with IL-17A and IL-17F gene polymorphism. In addition, BASDAI scores, family histories and HLA-B27 values were calculated with the allele frequency. Materials Methods: We examined 89 Ankylosing Spondylitis patients and 89 healthy controls. We looked IL17A(-197A/G) and IL 17F(7383A/G) polymorphism by using RFLP. Results: We found a significant relationships between IL17A(-197A/G) polymorphism and Ankylosing Spondylitis. There were no statistically significant relationships between the genotype distributions of IL-17A gene -197AG polymorphisms and BASDAI scores, and family history. However, there was a statistical relationship in the genotype distributions of patients, with HLA-B27 results. We couldn't find a relationship between IL-17F(7383A/G) and AS, BASDAI scores, family history and HLA-B27 results. Conclusion: In conclusion genetic and cytokine affect with AS has been studied per years. In our study there is a significant connection between AS and IL-17A gene polymorphism. This is the first study that found relationship between IL-17A gene polymorphism and AS. Further studies should be done to be sure for this relationship.
