Abstract (EN)
In this study, genomic libraries enriched with the repeats of `CA?, `GA?, `AAC? and `AAG? were developed by using DNA of pistachio cultivar `Siirt?. Selecting 57 clones from each library, a total of 228 clones were sequenced. Sixty-eight of these were duplicates, while 94 contained repeatative sequences and five contained duplicate sequences. Thus, primer development studies were attempted for the regions containing 99 repeatative sequences. Finally, eighty-four primer combinations were successfully designed. Among the sequenced clones, 36.8% of them generated primers. Among 84 primers tested, 59 generated (70.2%) bands. Among 59 primers generated, 13 primers were used to characterize 46 Pistacia genotypes belong to the 12 species. A total of 169 allelles were generated of which 165 were polymorphic. Average allel number per primer was 13.0, while average polymorphic allel number per primer was 12.7. Mean polymorphism information content of 13 primers were 0.905, while total resolving power was 5.39. According to Jaccard index beween 46 Pistacia genotypes changed from 0.08 to 0.98. The closest genotypes were `Ketengömlek? ? `Kırmızı? (0.98), and `Hacıreşo?- `Siirt? (0.98), while `PM-0104?- `Uzun? (0.08), `PM-0104?- `Ashoury? (0.08) and `PTX-0292?- `Ashoury? (0.08) were the most distant ones.
