Abstract (EN)
Anthemis pauciloba var. sieheana and Cota tinctoria subsp. euxina belonging to the family of Compositae were collected from Kayseri and İstanbul. The compositions of the essential oils of the aerial parts of the plants and fatty acids obtained from the hexane extracts were determined by using GC-MS. Within the scope of the thesis, antimicrobial and cytotoxic activity studies of these substances were carried out. In the essential oils of A. pauciloba var. sieheana 96 compounds were detected with 63.11%. It was understood that the main components of the essential oils are 1,8-cineole (8.499%), β-pinene (5.115%) and spathulenol (3.247%). In fatty acids, 6 compounds were identified with 97.115%. The main components were (Z,Z)-9,12-octadecadienoic acid (55.61%), hexadecanoic acid (12.831%) and octadecanoic acid (4.562%). In the analysis of the essential oils of C. tinctoria subsp. euxina, 84 compounds were determined with 74.94%. The main components were β-pinene (23.240%), α-pinene (6.518%) and limonene (3.655%). In fatty acids on the other hand, 7 compounds were identified with the yield of 81.342%. the main components were (Z,Z)-9,12octadecadinoic acid (72.353%), hexadecanoic acid (15.402%) and octadecanoic acid (4.936%). In order to determine the antimicrobial effects of the plants, it was tried to determine the minimum inhibition concentration values using different concentrations of essential oils isolated from the plants. The effects of the substances against different Gram-positive and Gram-negative bacteria were investigated and it was found that the essential oils of C. tinctoria subsp. euxina showed inhibitory effects against all bacterial strains at different concentrations. On the other hand, A. pauciloba var. sieheana showed inhibitory effect against Escherichia coli DH5α and Staphylococcus aureus at different concentrations. For the cytotoxic activity assay, HeLa cell line belonging to human cervical cancer was used. When cells were treated with different concentrations of hexane extracts, any cytotoxic effect was not observed.
