Abstract (EN)
ABSTRACT In this study the effects of different combinations of growth regulators on micropropagation of Aronia (Aronia melanocarpa (Michx.) Elliott) were investigated. The shoot tips in the production parcel of Isparta University of Applied Sciences, Faculty of Agricultural Sciences and Technologies were taken and planted in 5 different rejeneration and 5 different rooting environments. In this study the number of shoots, number of plants per explant, shoot length, vitrification percentage, number of nodes, rooting rate, number of roots per shoot and root length parameters were investigated. Key Words: Micropropagation, MS, BAP, GA3, Aronia 2019, 59 pages As a result of the data obtained, it is thought that the effect of different combinations should be examined together, not a single growth regulator, in order to determine the optimum environment in growth regulator concentrations. According to the data obtained, the medium for shoot propagation is medium containing the best number of plants per explant (4.56 number/explant), shoot length (20.55 mm/explant) and number of nodes (4.72 number/explant) MS + 1 mg / L IBA + 0.02 mg / L IBA + 1 mg / L GA3. The medium with the highest vitrification rate (%32.29) is the 5th medium with a concentration of 2 mg / L BAP. The best of ½ MS + 1 mg / L IBA in terms of number of roots (13,03), root length (23,26 mm) and rooting rate (%90,00). When the results of the study were examined, it was found that the use of very high or very low BAP and GA3 negatively affected the number of plants per explant. Rooting rates were low at both low and high IBA concentrations. In addition, it was determined that high BAP concentration increased vitrification rate in Aronia and thus negatively affected shoot development. In the light of all these results, this study reveals that the subculture numbers and the doses of plant growth regulators applied for propagation and rooting affects the results together. In the light of the data obtained, it was reveals that the effects of different combinations should be examined together in order to determine the optimum growth regulator concentrations. Therefore, it is considered that it is more appropriate to apply different plant growth regulators in the medium trials. As a result, the best medium for shoot propagation is medium containing 1 mg/L BAP + 0.02 mg/L IBA + 1 mg/L GA3. The best medium for rooting is medium containing ½ MS + 1mg/L IBA.
