Abstract (EN)

Aim: With this study we aimed to the effect of coenzyme Q10 and melatonin for Prevention hepatotoxicity induced by asetaminophen. In cases of hepatotoxicity, antioxidant effects of these substances should be tried and used to determine whether they can be used therapeutically and whether they have protective effects. In addition, the role of apoptosis in the mechanisms of hepatotoxicity will be evaluated. Material and method: The groups consisted of: In each group there were 10 Sprague Dawley rats (N=40). Group I (Control group), Group II (Acetaminophen group), Group III (Asetaminophen-Coenzyme Q10) and Group IV (Asetaminophen-Melatonin). Acetaminophen was single doze oral administration (1gr/kg). Melatonin treatment was given as intraperitoneal injections, 10 mg/kg each, at 30 minutes following acetaminophen administration. Coenzyme Q10 treatment was given as intraperitoneal injections, 10 mg/kg each, at 30 minutes following acetaminophen administration. The histopathological parameters were scored semi-quantitative using light microscope. For every slides related to each case in immunohistochemical (IHC) studies caspase3 and Bax antibody were used to cytoplasmic staining. The semi-quantitative scoring was done with 20 objective microscope. The ALT/AST measurement in blood serum was conducted with full-automatic biochemistr analyzer. The nitric oxide in blood serum was analyzed with ELISA method. Results: There were significant differences between the groups, control group and the group with acetaminophen in histopathologic parameters, IHC, biochemical serum levels of ALT and AST studies. There was no difference between serum NO of the groups. Besides, the mean of serum NO of groups were close and it was not found to be statistically significant (p>0,05). When groups with coenzyme Q10 and melatonin was compared, melatonin was found to have statistically significant impact on healing (p<0,05). Conclusion: Over doze of acetaminophen causes centrilobular necrosis. Heptatotoxicity was reversible and responded to treatment.When groups with melatonin and coenzyme Q10 were compared, melatonin was found to be protective and have more healing effects.
