Investigation of possible protective and therapeutic effects of curcumin and vitamin D in individual intestinal permeability in an experimental model of acetic acid induced ulcerative colitis

Abstract (EN)
Objective: This study was planned to investigate the effect of Curcumin and vitamin D on intestinal permeability in ulcerative colitis and the role of these agents in the inflammatory and oxidant process by looking at the levels of Occludin, one of the Zonulin and Tight Junction proteins, in an experimental ulcerative colitis model created with Acetic Acid. Material and Methods: Our study; Created from 48 Wistar albino female rats, 12 weeks old, weighing 250-300 grams; Control Group, Colitis Group, 200 mg/kg Curcumin + Colitis (Preventive treatment group), 0.4 µg/kg (16 IU/kg) Vitamin D + Colitis (Preventive treatment group), Colitis + 200 mg/kg Curcumin (Post treatment group), Colitis + 0.4 µg/kg (16 IU/kg) vitamin D (Post treatment group) was carried out with 6 groups of 8 rats. Interpretation of values and multiple comparison of group values; in Biochemistry with Dunnett's multiple comparisons test; In histology, it was performed with Dunn's multiple comparisons test. Results: In the control group, colonic TNF-alpha, IL-1β, IL-6, INF-gamma and MPO levels were found to be significantly lower than the colitis group; Colonic Occludin levels were found to be significantly higher than the colitis group. When the colitis group and the post and pre treatment groups are compared, it is observed that Colitis + Dvit TNF-alpha decreased; Colitis + Dvit OCC increased; Colitis + Cur and Cur + Colitis IL-1β decreased; Colitis + Cur and Cur + Colitis IL-6 decreased; Colit + Cur, Colit + Dvit and Cur + Colit INF-gamma decreased; It was observed as a result of the analysis that MPO decreased in all groups (p <0.05). In histopathological scoring, compared to the Colitis group, it was found that the most significant improvements in the parameters examined (except mucosal damage / necrosis) were realized in the group Colitis + Dvit (p <0.05). Conclusion: The results of our study prove the therapeutic and prophylactic efficacy of Curcumin through its anti-inflammatory and antioxidant mechanism. However, no significant conclusion could be obtained regarding the effect of Curcumin on intestinal permeability, but vitamin D is thought to have an important role in treating colitis-increased intestinal permeability through anti-inflammatory activity and lowering oxidant level.
