Study of biochemical activity characterization and isolation of secondary metabolites from lemon pulp

Abstract (EN)

In this thesis, compound structures of secondary metabolites which seperated and purified with chemical isolation method from lemon pulp which formed to be a by-product in juice factory were determined by NMR techniques. In addition, biochemical activities of certain components were determined. In this context, the pulp which are the result of the production of lemon juice were obtained from DIMES fruit juice factory and dried, was subjected to extraction by water boiling, the aqueous extract was extracted again with ethyl acetate to give ethyl acetate extract. This extract was fractionated into various fractions using different solvent systems by Sephadex LH 20 column chromatography, and was divided into several subfractions. The main components present in the extract; limonin, scopoletin, citropten, umbelliferon, hesperidin, hesperetin, catechol, benzenetriol, p-coumaric acid, prothocathecuic acid, diglikolic anhydrite, 2-hidroxy-4-metoxy benzenepropanoic acid methyl ester, 2-hydroxi-4-metoxy benzenepropanoic asit and eriodictiol were isolated with isolation processes. Antioxidant, reducing power, linoleic acid peroxidation inhibition, hydrogen peroxide removal and metal chelating activity tests on limonin and ethyl acetate extracts were found to be highly activities. From the isolated components, limonin, protocatechuic acid, eriodictiol and diglycolic anhydrite was tested in vitro against in PC 3 cancer cell lines. Within the scope of thesis study, chromatographic methods were developed that for separation, isolation and characterization of pure bioactive components from lemon pulps to use and evaluation as an alternative source.
