Researching the effect of avenanthramide C on breast cancer

Abstract (EN)

Aim: The formation of breast tumor occurs as a result of exposure to high levels of oxidative stress and free radical production. Antioxidant substances that can act against increased oxidative stress and free radicals can provide therapeutic efficacy in cancer treatment. In this study, it is aimed to evaluate the effects of Avn C that a substance with antioxidant origin, in breast cancer in vitro. For this purpose, it is aimed that Avn C could be a source for antioxidant-based treatment strategies. Material and Method: This study was performed on MDA-MB-231 and MCF-7 cells from breast cancer cell lines. MTS method, which provides information about cell viability to see the antiproliferative effects of Avn C, apoptosis analysis and Trypan blue staining methods for cell death studies, cell cycle analyzes for control cells and Avn C treated cells in both cell lines analyzed separately. After treating the cells with Avn C, clonogenic tests were performed to determine the colony forming abilities of the cells. In addition, invasion analyzes were performed to observe the effects of Avn C on invasion. Results: It was determined that Avn C showed activity in both cell lines when it was applied to MDA-MB-231 and MCF-7 cells from breast cancer cell lines. It was observed that it decreased cell viability in both cell lines with its antiproliferative effect and caused cell death with its cytotoxic effect. In addition, it was concluded that MDA-MB-231 cell line has limiting effects on cell cycle and also showed inhibitory activity against invasion of cells by invasion analysis for the same cell line. Conclusion: With this thesis, it can be suggested that Avn C compound may have therapeutic effects on breast cancer due to its antiproliferative effects, cell death triggering activities, cell cycle limiting and invasion inhibitory characteristics in breast cancer cells.
