Abstract (EN)

In this thesis, germination research of Barlia robertiana (Loisel.) Greuter salep orchids was carried out on the in vitro conditions. Barlia robertiana plants need mycorrhizal fungi to maintain their generations. Also, it takes many years to develop new mature plants. Barlia robertiana has a very high commercial value because salep is obtained from its tuber. In an attempt to obtain salep, this species are crafted from the nature insensibly and it endangered the continuity of its generation. In this study, two different concentrations (25% and 50%) of sodium hypochlorite solution were applied for seed sterilization. MS, ½ MS and OAM nutrient media with combination of Kinetin, BAP and NAA growth regulators at different doses in totally 6 medium were used and its effect on germination of Barlia robertiana seeds has been investigated. Barlia robertiana seeds were incubated four different photoperiod application which is light environment, 1 month dark environment, 2 months dark environment and 3 months dark environment and germination of seeds and their were investigated. In the study, the highest number of seed germination and protocorm formation were obtained from ½ MS + 4 mg / l Kin + 0.5 NAA medium. The best result was obtained from 50% sodium hypochlorite and fully lighted environment with 19 protocorms. The most germinated and most nutrient media were found to be ½ MS + 4 mg / l Kin + 0.5 mg/l NAA but applied applications are different. The highest germination and ability to form protocorms was observed in the seeds treated in the dark for 3 months.
