Somatic embryogenesis, histological analysis and investigation of synthetic seed production in some colchicum species

Abstract (EN)

In this thesis, production of synthetic seed and establishment of optimum regeneration protocol for somatic embryogenesis in C. cilicicum, C. baytopiorum and C. sanguicolle species were aimed. Corms of Colchicum species were used as explants. Different conncentration of 2,4-D (2, 3, 4mg/l) for C. cilicium and combination of NAA (0.5 ve 1 mg/l) ve TDZ (0.25, 0.5, 0.75 mg/l) for C. baytopiorum and C. sanguicolle were used. Swelling was shown in all species, callus growth was shown only in C. baytopiorum and C. sanguicolle. C.cilicium showed direct somatic embryogenesis. The highest embryo formation was obtained from RO medium with 4 mg/l 2,4-D for C.cilicium as 30%, from MS medium supported with 1 mg/l NAA ve 0.5 mg/l TDZ in C. baytopiorum as 35%. While cormlet formation was obtained from MS medium with 2 mg/l 2,4-D ve 0.5 mg/l 2iP in C. cilicium as %81.8, germination was observed from MS medium without hormone in C. baytopiorum as 14.7%. Histological analysis of embryos obtained was performed and synthetic seeds were produced.
