Abstract (EN)
It is very important to obtain strains that can be used in the production of industrial products from the isolates obtained by microbial diversity studies. Habitats in thermal areas, which are among the unique biodiversity of our country, are important in this sense. Within the scope of this thesis, thermal water samples taken from Çitgöl thermal springs in Simav district of Kütahya; Microorganisms thought to have the potential to exhibit protease activity were isolated by conventional microbial techniques. Species identification was carried out by DNA sequencing analysis of microorganisms, which were distinguished according to their morphological characteristics. From the Çitgöl thermal springs; Pseudoxanthomanas taiwanensis, Tepidomomas sp., Bacillus subtilis, Bacillus sp. and Brevibacillus borstelensis species were determined. The protease activity and protein amounts of these determined species were determined. Labeled as T5 as a result of the studies; Brevibacillus borstelensis was found to have the highest protease activity value. Protease produced by Brevibacillus borstelensis; It has been determined as an alkaline and thermostable protease producer due to its highest activity at 50°C and pH 12.
