The effects of some gums on the development of Bacillus coagulans and biofilm formation

Abstract (EN)
In this study, the effects of some gums (locus, pectin, xanthan and aljinate) which have prebiotic features on growth rate of B.Coagulans and biofilm is examined. While Microtiter plate method was used for biofilm formation, nutrient agar spreading method was used for observing growth rate of B. Coagulans. The absorbance value of biofilms of B. Coagulans was 0.11 measured at 630 nm by adding 0.4% crystal violet. The absorbance value of biofilms of B. Coagulans >0.1 indicates biofilm formation. Results obtained from studies to determine the effects of various gums (locust bean, pectin, xanthan, aljinate) on probiotic B. coagulans biofilm, The absorbance value measured at 630 nm with the addition of 0.4% crystal violet at the ratios of 2, 3, 4% respectively 0.27, 0.33 and 0.40 for the pectin gum, 0.44, 0.25 and 0.46 were measured for the xanthan gum, 1.68, 0.21 and 0.91 for the locust gum, 0.35, 0.32 and 0.26 for the aljinate gum. As a result of biofilm experiments, it has been shown that locust bean, xanthan, aljinate and pectin gums increase the biofilm-forming property of B. coagulans. While the initial cell count of probiotic B. coagulans used in the study was 7.69 log cfu / mL, it was determined that various gums increased the number of B. coagulans at the end of incubation. As a result of the addition of pectin gum, an increase of 0.22 log cfu / mL, an increase of 1.02 log cfu / mL and an increase of 1.51 log cfu / mL occurred at the rates of 2, 3, 4%, respectively. As a result of the addition of xanthan gum, a decrease of 0.27 log cfu / mL, an increase of 0.51 log cfu / mL and an increase of 2.26 log cfu / mL were observed at the rates of 2, 3, 4%, respectively. As a result of the addition of locust bean gum, an increase of 0.35 log cfu / mL, an increase of 1.06 log cfu / mL and an increase of 1.18 log cfu / mL occurred at the rates of 2, 3, 4%, respectively. As a result of the addition of aljinate gum, there was an increase of 0.41 log cfu / mL, 1.5 log cfu / mL and 1.78 log cfu / mL, respectively, at the rates of 2, 3, 4%. This increase showed that all used gums had a positive effect on B. coagulans bacteria growth.
