Identification, isolation and molecular characterization of phytophthora species from crown roted roots of some fruit trees

Abstract (EN)
Suspicious plant materials with Phytophthora spp. have been collected from some pome and stone fruits showing gummosis, crown rot, and brown fruit rot in 10 different countrysides and locations of Adana and Mersin provinces to identify characterization of Pythophthora species during 2009-2012. Phytophthora isolates from infected tissues of roots of apricot, apple and peach, and lemon fruits were cultured and purified on modified PARPH and PDA media including different concenrations of antibiotics in vitro condition. Colony patterns of Phytophthora isolates were performed on PDA, HA, KTA, YUA and MUA media and identified of their microscobic structures. Although, in general colony patterns of fungi isolates shown to be varied based on culture media, chrysanthemum for apricot isolates, stellate, camellia and chrysanthemum for apple isolates, uniform and camellia for peach isolate, radial, chrysanthemum, and rose petal for citrus isolates were created as colonial patterns. DNA fragments between 900-1050 bp of ITSI, 5.8S and ITSII regions of rDNA of Phytophthora isolates have been amplified by PCR and obtained restriction ragment length polymorphism by digesting with MspI and HaeIII seperately. Based on this digestion, citrus isolate, Phytophthora citrophthora as control has shown completely different DNA plymorphism from other isolates. The isolates APe-1 and MAş-1 have been considered as P. cactorum and P. megasperma rspectively. The isolate MYk-2 has been found some similarities to P. palmivora.Keywords: Colony pattern, ITS regions, PCR, Phytophthora spp., RFLP
