Optimization of plant regeneration from root explants at some citrus rootstocks

Abstract (EN)

In this thesis, root explants; Citrus sinensis L Osb. xPoncirus trifoliata L Raf. (cv `Carrizo Citrange?); Citrus aurantium L. (cv `Tuzcu 31-31?); Citrus aurantium L. (cv `Gou Tou?); Citrus reshni Hort. Ex. Tan.(cv `Tuzcu Cyprus Cleopatra Mandarin?) obtained from germinated seeds of four different citrus rootstock were used to optimize plant regeneration from root explants. Root explants belonging to citrus genotypes used in the present study were cultured in media supplemented with 21 different concentration of BA and NAA. From the results of study, the highest value of callus was determined from MS medium supplemented with 0.5 mg/L BA. in Carrizo Citrange 90.73%, in Tuzcu 31-31 Sour Orange 88.33%, in Tuzcu Cyprus Cleopatra Mandarin 62.87%, in Gou Tou Sour Orange 58.33%. The highest quality callus rates were observed and the highest success was obtained in Carrizo Citrange with 88.09%, and then in Tuzcu 31-31 with 25.95%. The lowest rate was determined in Gou Tou Sour Orange with 2.75%. No any observed in Tuzcu Cyprus Cleopatra Mandarin. In view of shoot formation rates, Carrizo Citrange identified as the most successful genotype (60.95%). In Carrizo Citrange, MS media supplemented with 0.5 mg/L BA, 1 mg/L BA+0.5 mg/L NAA ve 2 mg/L BA showed 100% success based on the shoot formation. In Gou Tou Sour Orange, shoot success rate was determined as 0.23%.
