Abstract (EN)
While the validity of innovative approaches in the field of drug discovery is increasing day by day, new studies are carried out to develop drugs with anticancer properties. Within the scope of this thesis, the cytotoxic effects of newly synthesized Palladium (Pd(II)) and Copper (Cu(II)) compounds were investigated by molecular docking on DNA molecule, cleavage on pUC19 plasmid DNA and cytotoxic effects on HCC1428 cell line by MTT assay. It was determined that these new compounds made intermolecular bonds with the DNA structure in docking simulations, caused DNA damage and fragmentation at certain concentrations in DNA cleavage experiments on pUC19 plasmid, and had a significant cytotoxic activity at all doses and incubation times studied with MTT assay performed on HCC1428 cells. Parallel to these cytotoxic activities, IC50 values were calculated. In DNA cleavage experiments, Cisplatin and Carboplatin anticancer agents were used as controls and similar results were obtained on DNA, and bands formed as a result of fragmentation were visualized by agarose gel electrophoresis technique. In the molecular docking simulations of the compounds, the binding affinities were found to be high and the images at the time of binding are shown as figures.
