ABSTRACTMSc THESİSIN VITRO GENOTOXIC EFFECTS OF BENZOYL PEROXIDE INHUMAN PERIPHERAL LYMPHOCYTESArzu YAVUZDEPARTMENT OF BIOLOGYINSTITUTE OF NATURAL AND APPLIED SCIENCESUNIVERSITY OF ÇUKUROVASupervisor : Prof. Dr. Mehmet TOPAKTAŞYear: 2005, Pages: 132Jury : Prof. Dr. Mehmet TOPAKTAŞProf. Dr. Rüştü HATİPOĞLUAssoc. Prof. Eyyüp RENCÜZOĞULLARIThe aim of this study was to investigate the possible genotoxic effects ofbenzoyl peroxide, which used as bleaching agent in flour, by determining in vitrosister chromatid exchange (SCE), chromosome aberration (CA) and micronucleus(MN) tests in human peripheral lymphocytes. The cells were treated with 25, 50, 75and 100 µg/ml benzoyl peroxide for 24 h and 48 h treatment periods.Benzoyl peroxide generally induced SCEs, but this effect was foundsignificant only at the highest concentration (100 µg/ml) for 48 h treatment period inthe present study. Also benzoyl peroxide increased both percentage of abnormal cellsand number of chromosome aberrations per cell (CAs/Cell) at 75 µg/ml and 100µg/ml concentrations for 24 h treatment period and at all concentrations for 48 htreatment period in human peripheral lymphocytes. This increase was dose-dependently manner. In addition to this, benzoyl peroxide affected negatively bothreplication index (RI) and mitotic index (MI). However this effect was foundstatistically significant only for 48 h treatment period. Benzoyl peroxide generallyinduced micronucleus formation. Besides, this effect was statistically significant at75 µg/ml and 100 µg/ml concentration for 48 h treatment period when comparedwith control.Key words: Human Peripheral Lymphocytes, Benzoyl Peroxide, Sister ChromatidExchange (SCE), Chromosome Aberration (CA), Micronucleus (MN)II
