Optimization of ß-galactosidase catalyzed synthesis of galactooligosaccharides using response surface methodology

Abstract (EN)

In this study, it was aimed to synthesize galactooligosaccharide (GOS) from lactose by using Kluyveromyces lactis ß-galactosidase and to optimize the reaction conditions by using response surface methodology. 4-factor and 3-level Box-Behnken design was used for response surface methodology. The four independent factors for GOS synthesis were chosen as reaction pH, temperature, initial lactose concentration and reaction time. The highest GOS yield was obtained as 17,2 % (w/w) when the reaction pH was 6, temperature was 60 oC, initial lactose consantration was 300 mM (10% w/v) and reaction time was 6 hour among the 27 different sets of experimental conditions. It was determined that the usability of mathematical form of proposed model was suitable in the prediction of GOS yields of GOS synthesis catalyzed by using Kluyveromyces lactis ß-galactosidase.
