Effects of genotype and plant growth regulators on the haploid plant regeneration from anthers of field pea

In this study, it was aimed to determine the possibility of the obtaining haploid plants from the anthers of five field pea (Pisum sativum L.) cultivars cultured on the mediums containing MS minerals+B5 vitamins+20 g/l saccarose+20 g/l maltose+ 500 mg/l glutamine+ 20 ml/l coconut water+6 g/l agar and combinations of different concentrations (1 mg/l or 2 mg/l) of two different auxins (2,4-D and Picloram) and different concentrations ( 0, 0.25, 0.5 or 1 mg/l) of BAP. The results of the study showed that concentration of the BAP significantly affected the reaction rate of the anthers and 0.25 mg/l concentration of the BAP was optimal for the optimum anter reaction rate. Anther reaction rate was not statistically significant influenced by genotype, auxine type and concentration. In the experiment, averaged value of the anther reaction rate was determined as 3.9 %. Among the field pea cultivars tested, anthers of the cultivar Burdur showed highest anter reaction rate (6.6 %) while anthers of the cultivar Tekirdag showed the lowest anter reaction rate (2.4 %). The calli and embryoids from the anthers of the field pea cultivars tested did not regenerate any plant. From the results of the study, it was concluded that further research on the effects of the different stress treatments and different regeneration mediums must be conducted with the cultivars Burdur and Carina being most responsive cultivars in the study Keywords: Pea, Anther culture, Genotype, Auxine type, BAP, Concentration
