Abstract (EN)

Bioinformatics tools (in silico tools) can be used to identify allergens from different sources and to find the compounds that are associated with food allergens. Identification of allergen proteins by using wet-lab technology is a time-consuming and also costly process. Thanks to the development in the field of bioinformatics, it is now possible to estimate the allergen proteins. In the present thesis, four different computer-based comparison tools were used to investigate the allergic effects of potential allergen proteins in different plants and previously unknown to be allergens. For multiple sequence alignment Clustal W, for homology modelling SWISS-MODEL, for comparison of protein parameters ProtParam and for allergen estimation AlgPred were used. In addition, BIOPEP tool is used for in silico hydrolysis. Considering the results, seven proteins Corchorus olitorius, Cucumis sativus, Capsicum chinense, Carica papaya, Morus notabilis, Theobroma cacao and Jatropha curcas were defined as potential allergens in different species. At the same time, potential allergen proteins were analysed by BIOPEP. At the end of the analysis, the bioactive peptides in the potential allergen proteins and their functions were explained. Non-allergenic forms of potential allergen proteins were tried to be obtained with the results of in silico hydrolysis. In conclusion, in silico tools developed under the field of bioinformatics can provide a big contribution to the estimation of unknown allergen proteins in different fruits. Based on the in silico results, physicians can suggest people who have allergenicity to kiwellin not to consume the fruits that contain kiwellin-like proteins.
