Abstract (EN)

Collagen production in people's bodies has decreased at younger ages due to reasons such as living in a stressful and unhealthy environment, changes in food consumption habits and changes in food ingredients. In the meantime, the aging of the world population has led to the need to take collagen as a supplement. Collagen is obtained from different sources by different methods. This Master's thesis aims to obtain collagen from biological material using chemical and enzymatic methods and to increase collagen yield by using experimental statistical method. For this purpose, waste eggshell membrane was selected as the collagen source due to its high consumption, waste and easy supply of waste. Egg shell membranes were extracted using two different extraction methods (ultrasonically assisted enzymatic extraction and acidic extraction). Extraction temperature, sonication time and enzyme amount conditions were optimized by central composite design method during the extraction of collagen from eggshell membranes using ultrasonically assisted enzymatic extraction method. Highest yield and purity was found to be 0.95 and 19.1 µg/µL under the conditions of 0.5 g enzyme amount, 10°C extraction temperature and 45 min sonication time. The collagens obtained were subjected to FTIR analysis. It was observed that the FTIR results of the samples were similar to each other and their helical structures were intact.
