Determinaion of actinobacteria biodiversity of the Bolu Yenicaga Lake

Abstract (EN)
The identification of the biodiversity of Actinobacteria in Yeniçağa Lake and addition of possible novel species to the literature is purpose in this study. In this study, Actinobacteria were isolated from 10 different media according to dilution plate technique. As a result of the isolation, 92 isolates were obtained. 92 isolates were isolated 6 from SM2 agar, 25 from starch-casein agar, 12 from SM3 medium, 9 from Marine agar, 4 from Humic acid vitamin agar, 17 from M1 agar, 11 from Nocardia agar and 8 from R2A agar. 76 isolates were identified at the genus level by performing 16S rRNA gene region sequence analysis. Phylogenetic analyzes were performed using MEGA 7.0 and PHYDIT programs. As a result of this study 37 isolates belong to Streptomyces, 18 isolates belong to Nocardia, 6 isolates belong to Nonomuraea, 5 isolates belong to Micromonospora, 4 isolates belong to Actinomadura, and 1 isolates belong to Amycolatopsis, Cellulosimicrobium, Glycomyces and Oerskovia. Phylogenetic dendograms based on sequence analysis of 16S rRNA gene regions of the genus were examined and it was determined that 10 isolates could be novel species. 9 of these isolates belong to the genus Streptomyces and 1 belong to the genus Amycolatopsis. MLSA analyzes of 9 Streptomyces isolates were performed. It was determined that the phylogenetic distances of the isolates were higher than 0.007 with the obtained results. Differences between isolates and type strains were determined by performing numerical studies of possible novel species according to the MLSA and 16S rRNA gene region data. Streptomyces sp. YC419 and Streptomyces sp. DNA:DNA hybridization experiments were carried out with the type strain of YC504 isolates to assure that the isolates were the novel species. The morphological and cultural characteristics of isolates and the type strain were defined by inoculation to 10 different media.
