Özet (EN)

Aim: The aim of this study is to investigate the cytotoxic effect of borax pentahydrate (BPH), in this area new boron compound, on glioblastoma multiforme (GBM) cell line U-87MG, which is a new boron compound, and which cell death mechanism is triggered. Material and methods: The cytotoxic effect of BPH 'in U-87 MG cell line was measured by MTT assay. Then, a technically operated Muse ® cell analyzer with a fluorescent base was used in the direction of apoptosis and autophagia with the determined IC50 dose. The obtained data were evaluated using GraphPad prism statistics program. Results: As a result of cytotoxicity experiments, the IC50 value of BPH in U-87 MG was found to be 2.454 μM. In the apoptotic control group, 41% of cell deaths were found in the 3% test group (P <0.01), depending on the IC50 value. In the autophagy evaluation, the control group had a density value of 1.0 and the experimental group had a value of 1.2 (p> 0,01). Statistical evaluation of the findings showed a significant effect on apoptosis; autophagic direction. Conclusions: This was found to be a cytotoxic effect on the U-87 MG cell line modeling BPH 'in GBM under the light of this data. It has been determined that this effect is achieved via apoptosis mechanism instead of autophagy cell death mechanism. These results suggest that BPH may be an agent that can be used in the treatment of GBM which is resistant to apoptosis-stimulating chemotherapeutics.
