Abstract (EN)

Objective: Parkinson's disease is one of the most common neurodegeneration disorders in the world caused by the decrease in the amount of dopamine in the basal ganglia and the death of cells in the substantia nigra. Studies on growth hormone have shown that is an anti-apoptosis protein, GH has proliferative effect on neuron cells and increases the amount of dopamine. In this thesis, it was aimed to determine the neuroprotective effect of GH on SH-SY5Y cell line modelled with 6-OHDA in-vitro Parkinson disease. Material and methods: The morphological change in the cells was observed by administering the lethal dose of 6-OHDA with 100 µm / ml and 1000 ng / ml BH. Cell survival was calculated by MTS and live cell counting. The mRNA was isolated from cells and qPCR was performed with this. Thus, the possible effects of GH were also examined genomically. Results: It has been demonstrated that Growth Hormone can inhibit 6-OHDA-induced mitochondrial dysfunction and oxidative stress-induced cell death in neuronal cells. Thus, it protects the cells. Conclusion: This is the first study to demonstrate that GH can prevent apoptosis and have a proliferative effect in parkinson cell modeling which made by 6-OHDA. In the light of the findings obtained from the study, we think that GH may be a potential neuroprotective or therapeutic agent for parkinson's disease.
