Abstract (EN)

Brain cancer is a type of cancer that is difficult to diagnose and treat. In most cases, there is no response to chemotherapy and radiation therapy. For this reason, alternative therapies are being sought. Research is being carried out into natural treatment methods in Turkey, which is rich in species. The overall aim of this dissertation is to investigate the effect of Rhododendron Luteum plant on C6 glioma brain cancer and to develop a new treatment method. The Rhododendron Luteum plant, which has a flavonoid content, is used in the treatment of various diseases. The basic principle of this thesis is to determine the effect of the Rhododendron Luteum plant on the autophagy mechanism in the glioblasto cell. The MTT assay was performed to monitor the cytotoxic effect of Rhododendron Luteum extract on C6 glioma cells. The present study investigated the effects of varying 8 concentrations of Rhododendron Luteum extract (5, 10, 50, 75, 100, 125, 250, 500 μg/mL) on the proliferation and viability of C6 glioma cells. In addition to cytotoxicity assessment, total antioxidant and total oxidant property determination were studied using the same concentrations as the TAS/TOS test. According to the results of the study, the TAS value increased depending on the concentration doses used, while the TOS values decreased. Measurements were made in gene expression of plant extraction using RT-PCR Gene Expression method.
