The effect of cerebrolysin on cisplatin induced oxidative brain injury in rats

Abstract (EN)

Background and aim: Cisplatin is a platinum-derived anticancer drug that is widely used in the treatment of many cancer diseases. Anticancer doses of cisplatin cause serious damage to brain tissue. The role of oxidative stress and inflammation is known to be in the pathogenesis of cisplatin-related brain injury. Cerebrolysin is a neuroprotective and neurotrophic-derived drug with low molecular weight peptides and amino acids. Therefore, the aim of this study is to investigate the effect of cerebrolysin on cisplatin-induced oxidative brain injury in rats biochemically and histopathologically. Materials and methods: Albino wistar rats used in the experiment were divided into three group as healthy control group (SG, n=10), the group given cisplatin at a dose of 2.5 mg / kg (CIS, n=10) and the group given 2.5 ml / kg cerebrolysin + 2.5 mg / kg cisplatin (CCG, n=10). Cerebrolysin and cisplatin were injected intraperitoneally (ip). Distilled water was used as the solvent for CIS and SG groups. Cerebrolysin, cisplatin and volumetric water were administered in the same dose and volume once daily for 14 days by the same method. At the end of this period, the animals were sacrificed by high dose (50 mg / kg) thiopental anesthesia and brain tissues were removed. Malondialdehyde (MDA), myeloperoxidase (MPO), total glutathione (tGSH), superoxide dismutase (SOD), total antioxidant (TAS) and total oxidant status (TOS) levels were measured in the extracted brain tissues and tissues were examined histopathologically. Results: MDA, MPO and TOS levels were significantly higher (P <0.001) tGSH, SOD and TAS levels were lower (P <0.001) compared to cerebrolysin group. In the Cerebrolysin group, histopathological damage was minimal. Conclusion: Our results indicate that cerebrolysin may be useful in the treatment of cisplatin-induced oxidative brain injury.
