Peach (Prunus persica L. (Batsch)) is a plant cultivated as a fruit and ornamental plant. Conventional vegetative propagation methods of peach are often time consuming and have a low growth rate depending on the season. This allows for limited use of valuable plant materials. In recent years, it is being cultivated of fruit seedlings out by using biotechnological methods, which include in vitro culture methods and allows for high yields and disease-free plant production. In this study, sterilization, shooting, rooting and acclimatization studies had performed using nodal explants to develop a protocol for micropropagation of peach plant. Nodal explants had first subjected to surface sterilization; In commercial NaOCl, they had kept in different concentrations and at different times. As plant growth regulator for shoot formation (BBD); 0.5 and 1 mg / L 6-Benzylaminopurine (6-BAP) together with 0.25, 0.5, 1 mg / L of 3 different concentrations of 5 types of auxin [Indol-3 Acetic acid (IAA), Indol-3 butyric acid (IBA), Naphthalene acetic acid (NAA), Picloram (P), 2,4-Dichlorophenoxy acetic acid (2,4-D)] had used. MS (Murashige Skoog, 4.4 g / L) had used as the medium, 30 g / L sucrose, 8 g / L agar had added and the pH had buffered in the range 5.75-5.80 and had autoclaved for 15 minutes at 121 ° C. It had taken to the culture room at photoperiods of 16 hours light / 8 hours dark at 24 ± 2 ◦C. Observed shoot at certain intervals, shoot, the number of leaves and length had recorded. Data had subjected to analysis of variance (ANOVA). The best sterilization, had obtained in the study in which the undiluted commercial NaOCl had treated for 13 minutes. The ability to form shoots had observed on the medium containing the best 1 mg / L BAP + 1 mg / L NAA. Only callus formations had observed instead of shoot in medium containing "P". The minimum shoot formation had recorded on media containing 2,4-D. Rooting stage; MS + Sucrose and 0.25 0.5, 1, 2 mg / L IBA, NAA, 2,4-D, IAA had used in 6 different protocols. The highest rooting had recorded on 1 mg / L NAA medium containing ¼ MS + ¼ sucrose. No rooting had not been on medium containing IAA. In the last stage, rooted plantlets which have completed their development were taken into acclimatization environments and survival rates were calculated. Key Words: Prunus persica (L.)
