Identification of bacillus species isolated from different retail meat, investigation of their proteolytic and lipolytic enzymes activities

Abstract (EN)

In this study, the prevalence of Bacillus species and the number of coliform group microorganism / E. coli was investigated a total of 45 samples of meat products consisting of 15 meat balls, 15 minced meat, 15 chicken meat collected from different butcheries and retail markets in Ankara. The proteolytic and the lipolytic activities of identified this Bacillus species was studied. At the same time in this study the antimicrobial activity of essential oil that obtained from Satujera wiedemanniana (Lalem.) Velen. against Bacillus species was investigated. Bacillus species were detected on all of analyzed 15 meat balls (%100) and 15 minced meat samples (%100) and 12 of 15 chicken meat (%80). From this samples 279 isolates were obtained. This isolates were identified as 121 (%36,2) B. circulans, 52 (%18,6) B. firmus, 40 (%14,3) B. lentus, 39 (%13,9) B. megaterium, 34 (%12,1) B. licheniformis, 9 (%3,2) B. mycoides, 3 (%1) B. sphaericus and 1 (%0,3) B. cereus. B. circulans were the most isolated Bacillus spp. from meat balls and minced meat samples 42 (%63,6), 52 (%29,5) respectively, and 9 (%24,3) B. megaterium were the most isolated Bacillus species from chicken meat. In the examined samples, the number of coliform group microorganism / E. coli was calculated by using Most Probable Number technique. It was found vii between <0,30 unit/gr and >110,0 unit/gr. The number of coliform group microorganism samples were counted in 24 samples >110,0 unit/gr. The number of E. coli were found in 5 samples >110,0 unit/gr. The proteolytic and the lipolytic enzyme activities of identified Bacillus isolates were evaluated. The proteoliytic enzyme activity of 168 of 279 (%60,2) Bacillus isolates and the lipolytic enzyme activity of 132 of 279 (%47,3) Bacillus isolates found as positive. The antimicrobial activity of essential oil obtained from Satujera wiedemanniana (Lalem.) Velen against Bacillus species was investigated by the agar-well diffusion method. Essential oil showed the strongest antimicrobial activity against B. megaterium (17,0±3,0 mm), while showed the weakest antimicrobial activity against B. cereus (3,0±0,5 mm).
